We have implemented or developed the following technologies to accomplish the goals of our Human Brain Project: One and two color dye injections into neurons and glia in fixed slices. Combination of dye injection with immunocytochemistry to investigate protein distribution in identified neurons Tomographic reconstruction of spiny dendrites in thick slices using the 400 keV intermediate voltage electron microscope and NCMIR and the 3 MeV ultrahigh voltage electron microscope at Osaka University. Correlated light and electron microscopic 3D datasets 3D protein localization at the light and electron microscopic levels using confocal microscopy and electron tomography. An Oracle-based database for 2D and 3D confocal and electron microscopic datasets. A high-resolution, custom designed 2K X 2K digital camera to acquire tomographic datasets Remote operation of our specialized IVEM through a web-based interface to acquire datasets for tomographic reconstruction.
