Three-dimensional structure & function of synapses in the brain

Software tools have been developed to reconstruct three-dimensional structure from serial section image data on a Microsoft Windows platform. Tools are available to align serial sections, to page through serial sections, to identify and trace the outline of objects that traverse the sections, and to reconstruct these objects in three dimensions. These tools are generally applicable to all sectioned tissues, whether they were imaged by light microscopy, confocal microscopy, or EM. A database of ultrastructural brain anatomy is being compiled and put into a range of formats appropriate for WWW access. Online data includes segments of spiny dendrites of CA1 pyramidal cells taken from stratum radiatum, and high magnification 3D data on individual spines and their synapses. We are regularly adding new synapses and dendrites to this database. In addition, there are tutorials on how to identify specific structures on electron micrographs from the brain neuropil.
